Would a 3/8" brass bolt be big enough to carry 300 amps? It will be used to clamp down connectors in a junction box I am building.

If you plan on using 300 amps on a continuous basis, you better add a couple more alternators. Short term, such as a large on board welder with electric start, a 3/8ths bolt should be fine.
 
More importantly;

when you use the bolt to make the connection, It would seem you will probably have a lug to slide under the bolt?... make sure you solder the lug to the wire. no crimping allowed at these current levels.

Use an external or internal aggresive star washers as well.

Bolting a flat ring lug to a flat surface under the bolt head results in a connection that can become oxidized and suffer from increased resistance over time.
 
the standard electronic solder... 63/37 rosin core flux solder is fine.

you can pick it up a Radio Shack, pick up some tuner cleaner to clean the flux off of the solder joint when your done. You can use an old tooth brush to scrub the solder joint clean.
