I have a 2000, 2500 4x4 and am going to change the front and rear diff oil. If I use Amsoil Long Life 75W-90 Gear Lube, do I need to add the slip-lock to it or is it ok by itself?

I have done the same and not added any slip it.

I do not have any chatter, and the posi works great.

... however mt rear tires are wearing a little bit quicker than my fronts... but that is normal... probably no relation.
 
Been using straight Amsoil 75W90 with NO additives in my 5-spd and rear differential for approx 220000 miles - no complaints whatsoever.

One thing to consider is how YOU use your truck. I, for instance, do NO towing and most of my miles are on longgg, flat, highways in SouthEast Texas.

I put 75W90 in the front and 75W140 in the rear, doing the figure 8's in a parking lot showed that I need to add a little modifier, a couple years later, I had to add a little more. Used the 75W140 in the rear because I tow a 11K 5er. SNOKING
 
I had a similar experience as Snoking. Was fine for about a year then started to chatter. Added some slip modifier and still chatter! Must be time to change the lube.
