Just purchased 10 qts of Mopar 04874464AB for my next fluid change (8 qts + 2 qts for 'Fast Coolers' from Geno's). With this OEM fluid, do I need to add the Mopar friction additive, or is it already in the bottle?

I thought the NV5600 took Pennsoil Syncromesh and I've never heard of adding friction modifier to a transmission.
 
No additive needed. The factory fill is 4 qts, plus coolers, maybe 6 total. I would not dump 10 qts in, you will blow your seals and cause heat.

I was in a local diesel shop a couple of weeks ago, and they told me to add the trans oil to full then pull the top cover and add 1 more quart. Seems like a lot. Also, what about just using 50wt synthetic oil?

That might be okay because it only takes 4 qts to the fill plug, so adding another qt makes sense if the book calls for 9.5 pints. I am thinking that NV used that fill quantity with the transmission sitting level on a bench. In the truck, with the angle it is mounted at it takes less. I change mine every 36k so that means I have changed it 7 times and I can only get 1 gallon in. Maybe I will add an extra pint of so the next time I change it.

I fill my nv5600 to the fill plug, then I use one of the small pumps that attaches to the oil bottle. I then use a chunk of clear vinyl hose that will fit in the PTO cover bolt holes. I pump in fluid until it comes out the top PTO bolt hole. With Blumenthol covers, that is about 9qts.
