I have a 98 12 valve w/ the NV4500, right now is stock. I would like to get to 400-450 RWHp. Fuel mileage and controlling EGTs while towing (livestock) are my main concerns. I was thinking about BD twins, DDP Stage 4s, a #10 plate, and a SBC Con OFE. My questions are:

1. Would this be a good combination?

2. Can I achieve this level w/o compromising my headgasket? (boost pressure)

3. What kind of timing should I give it?

I would go with a conFE and maybe some smaller injectors. The combo you have listed would put you well into the high 4's/low 5's. You would have to keep the plate back to be around the low 4's. Headgasket is questionable, the BD twins do 65PSI with no problem especially under load. look into some 12MM headstuds at a minimum.
 
Would sliding the plate back w/ bigger injectors be worse than plate forward w/ smaller injectors?

Can I adjust the wastegate on the twins to help keep head gasket alive? (I still think I will do 12mm studs. )
 
Nah I wouldnt say worse, you can go with the 4's and keep the plate back. Thats how I have mine setup now and its defenetly driveable. One thing with big injectors is there always there, where with smaller ones with a plate you can play with what you want for fuel delivery. DDP3's are like 370 marines, but with the proper spray pattern. the 4's are about 60hp over those. Ross at DDP is a easy person to talk to if you want to pick his mind.

With either choice injector and your pump, your power goals are defenetly attainable.
