I'm considering purchasing an '01 2500 4X4 SWB 6sp with about 159,000 miles and I was wondering what issues I need to be aware of with this setup.

I ran one & will have it back on the road again soon. It's a great combination of ingredients. If you do nothing else to it, get gauges & keep tabs on the fuel pressure. the lift pump is a weak link & the clutch won't handle much more than stock power.
 
I recently sold a truck just like yours. Darkhorse nailed it. The stock clutch can only hold stock power and the liftpumps are known to have issues. It is really hard to get the parts to convert the towing mirrors to power if desired.
 
... i just traded mine ,i had 236k on it,as mentioned earlier... . add gauges for boost/egt and fuel pressure..... i used a FASS 95gph instead of the stock lift pump..... worked GREAT... . the stock clutch would hold with RV 275 injectors,but as soon as i added an EDGE EZ i had to go with a new SB Con OFE... . another winner in my book!!! KEEP AN EYE ON YOUR TRANSMISSION FLUID!!!... . thats what i DID NOT do,and i payed the price..... scratch 1 nice transmission!!!!..... the 275's and EZ dynoed 312 RWHP... . which made towing my 5'er a breeze..... good luck with your purchase!!!... ... tom:)
 
track bar and steering are weak spots, too. i recently installed a Thuren crossover steering set up as well as his track bar and the difference is HUGE! i also agree with all of the other responses listed.
